dc.description.abstract |
Zubní sklovina je nejtvrdší tkáň v těle obratlovců. Z 98 % je tvořena minerálem hydroxylapatitem uspořádaným do podoby tzv. prizmat obklopených aprizmatickou hmotou. Sklovina vzniká procesem biominaralizace jako sekreční produkt buněk ameloblastů. Tyto speciální buňky produkují během amelogeneze velké množství proteinů, mezi které patří především amelogenin, ameloblastin, enamelin, tuftelin a mnohé další. Tyto proteiny poté slouží jako nosiče pro hydroxylapatitové krystaly a jsou nezbytné pro jejich další vznik, růst a formaci. Kromě toho mají i další biologicky významné funkce. Cílem této práce bylo připravit ameloblastin divokého typu a jeho mutant postrádající část kódovanou exonem 5 v expresním systému E. coli a optimalizovat jejich purifikační protokol. Dalším cílem pak bylo otestovat, zdali mají tyto proteiny antimikrobiální vlastnosti. |

Tooth enamel is the hardest tissue of the body of vertebrates. It is formed by 98 % of hydroxylapatit mineral organized in so-called rods surrounded by interrod substance. Enamel is formed by process called biomineralization as a secreted product of ameloblasts. During amelogenesis these special cells produce a great amount of proteins, especially amelogenin, ameloblastin, enamelin, tuftelin and many others. These proteins are used as scaffolds for hydroxylapatite crystals and they are necessary for further nucleation, growth and formation of these crystals. Moreover they have many other biologically important functions. The aim of this thesis was to prepare wild type ameloblastin and its mutant without the part encoded by exon 5 in expression system of E. coli and to optimize their purification protocol. Next aim was to test potential antimicrobial behaviour of these proteins. |
