dc.description.abstract |
Tato diplomova prace je zaměřena na využiti disperzni kapalinove mikroextrakce ve spojeni s kapalinovou chromatografii s UV detekci pro stanoveni alfa- a beta-hořkych kyselin v klasickem pivu a v pivu chmelenem za studena. V teoreticke časti jsou popsany různe způsoby modifikace disperzni kapalinove mikroextrakce, možnosti spojeni teto mikroextrakce s kapalinovou chromatografii a dalšimi technikami. Zaměřena je i na vyrobu piva, popis různych způsobů chmeleni a stanoveni latek obsaženych v pivu, zejmena těkavych latek a hořkych kyselin. V experimentalni časti byly nejprve optimalizovany podminky 4 disperznich kapalinovych mikroextrakci využivajici ztuhnuti organicke kapky (DLLME-SFO, AA-LLME-SFO, UA-LLME-SFO, UA-DLLME-SFO), ze kterych byla vybrana nejvhodnějši mikroextrakce pro stanoveni hořkych kyselin. Dale jsou prezentovany vysledky analyzy realnych vzorků piv a porovnani množstvi hořkych kyselin v pivech chmelenych klasicky a pivech chmelenych za studena. |

This thesis deals with the use of dispersive liquid-liquid microextraction followed by liquid chromatography with UV detection for the determination of alpha- and betta-bitter acids in classical beer and in dry-hopped beer. In the theoretical part there are described various modifications of dispersive liquid-liquid microextraction methods together with possibilities of their connection with liquid chromatography and other techniques. It is also focused on the production of beer, the description of the different ways of hoping and the determination of substances in beer, especially volatile compounds and bitter acids. In the experimental part, the conditions of the 4 dispersive liquid-liquid microextraction methods with solidification of floating organic drop (DLLME-SFO, AA-LLME-SFO, UA-LLME-SFO, UA-DLLME-SFO) were optimised and the most suitable approach for the determination of bitter acids was selected. In addition, the results of the analysis of real beer samples and the comparison of bitter acids in classical beer and dry-hopped beer are presented. |
