Cholinesterasy (ChEs) jsou skupinou enzymů, které katalyzují hydrolýzu acetylcholinu na cholin a kyselinu octovou. Jedná se o základní proces umožňující obnovu cholinergního receptoru. Mezi skupinu esteras se řadí acetylcholinesterasa (AChE) a butyrylcholinesterasa (BChE). Oba enzymy mají z 65 % stejnou sekvenční homologii aminokyselin a jsou kódovány různými geny na lidských chromozomech. Tyto dva enzymy se navzájem liší substrátovou specifitou a místem exprese. Inhibitory cholinesteras (galantamin, donepezil a rivastigmin), které zvyšují cholinergní neurotransmisy, jsou využívány pro snížení příznaků Alzheimerovy choroby inhibitory Brutonovy tyrosinkinasy (BTK) (tirabrutinib, evobrutinib a branebrutinib) snižují aktivaci fosfolipasy γ 2, která byla identifikována jako genetický rizikový faktor Alzheimerovy choroby. |
