dc.description.abstract |
Disertační práce se zaměřuje na popis apoptózy a na přehled bioanalytických
metod, které se využívají při její detekci a hodnocení. V rámci disertační práce jsme
s využitím sondy Hoechst 33258 vyvinuli spektrofluorimetrickou metodu pro detekci
nukleární kondenzace a fragmentace. Pomocí této metody jsme popsali vliv
modelových toxinů na proces nukleární kondenzace in vitro. Následně jsme výsledky
získané novou metodou porovnali s výstupy dalších metod určených k detekci
apoptózy, tj. detekce aktivity kaspáz, JNK a PARP aktivace, a metody TUNEL a DNA
žebřík. V závěrečné části práce jsme naměřené výsledky shrnuli, diskutovali
a porovnali s literaturou. Nově vyvinutá metoda využívající Hoechst 33258 je plně
porovnatelná s metodou TUNEL a má navíc řadu výhod (např. rychlost, kvantifikace,
nízká cena). Další velkou výhodou je možnost analyzovat velké množství vzorků
najednou. |

This doctoral thesis focuses on explaining the process of apoptosis and on an
overview and description of bioanalytical methods used in its evaluation and detection.
In the experimental part of this thesis, we developed the spectrofluorometric method
Hoechst 33258 for the detection of nuclear condensation with subsequent evaluation.
Using this method, we detected the effect of selected toxins on the process of nuclear
condensation in vitro.
Subsequently, we compared the results obtained by the new method with the
outputs of other methods designed to detect apoptosis, ie detection of caspase activity,
JNK and PARP activation, and TUNEL and DNA ladder methods.
In the final part of the work, we discussed the results and compared them with
the literature. Newly developed method using the Hoechst 33258 probe is fully
comparable with TUNEL method. Moreover, our method has a number of advantages
(e.g. speed, quantification, low cost) over the TUNEL assay. Another great advantage
is the ability to analyze a large number of samples at once. |
