dc.description.abstract |
Oxidační stres vzniká v důsledku nerovnováhy mezi vznikem a odstraňováním reaktivních forem kyslíku a dusíku. Působením volných radikálů na lipidy dochází k procesu lipoperoxidace. Jako nejvýznamnější sekundární produkt vzniká 4-hydroxynonenal, který se podílí na rozvoji řady onemocnění a je považován za marker oxidačního stresu. Symetrický a asymetrický dimethylarginin vznikají methylací argininu působením enzymů ze skupiny protein-arginin-methyltransferáz a jsou degradovány pomocí enzymu dimethylarginindimethylaminohydrolá-zy. Oxidační stres způsobuje zvýšení exprese protein-arginin-methyltransferáz a naopak snížení aktivity dimethylarginindimethylaminohydrolázy, což vede ke zvýšení hladin asymetrického dimethylargininu. Asymetrický dimethylarginin inhibuje aktivitu syntázy oxidu dusnatého, což vede k endoteliální dysfunkci a prohlubuje oxidační stres. K nadprodukci volných radikálů dochází např. vlivem vysoké hladiny glukozy. Produkty oxidačního stresu následně přispívají k rozvoji diabetu mellitu, 4-hydroxynonenal vyvolává apoptozu -buněk Langerhansových ostrůvků pankreatu, asymetrický dimethylarginin způsobuje endoteliální dysfunkci a s ní spojené kardiovaskulární či cévní onemocnění. |

Oxidative stress is formed from an imbalance between the formation and removal of reactive forms of oxygen and nitrogen. The effect of free radicals on lipids causes the lipoperoxidation process. The most important secondary product is 4-hydroxynonenal, which is involved in the development of many diseases and is considered a marker of oxidative stress. Symmetric and asymmetric dimethylarginine is formed by arginine methylation by enzymes from the protein-arginine-methyltransferase family and is degraded by the enzyme dimethylarginindimethylaminohydrolase. Oxidative stress causes an increase expresion of protein-arginine-methyltransferase and then inhibits the activity of dimethylarginindimethylaminohydrolase, resulting in increased levels of asymmetric dimethylarginine. Asymmetric dimethylarginine inhibits the activity of nitric oxide synthase, resulting in endothelial dysfunction and increased oxidative stress. Overproduction of free radicals occurs, for example, due to high glucose le-vels. Oxidative stress products subsequently contribute to the development of diabetes melli-tus, 4-hydroxynonenal induces apoptosis -cells of Langerhans' pancreatic islets, asymmetric dimethylarginine causes endothelial dysfunction and with it associated cardiovascular and vascular dinase. |
