| dc.description.abstract |
Tato diplomová práce se zabývá stanovením derivátů flavonu a methylxantinu v nápojích obsahující čajový extrakt. Nejprve byly optimalizovány podmínky separace metodou kapalinové chromatografie s UV detekcí a následně metoda přípravy vzorku. Pomocí optimalizované metody HPLC-DAD byly stanoveny jednotlivé derivátu flavonu a methylxantinu ve vzorcích ledových čajů obsahující extrakt zeleného nebo černého čaje, v sáčkových zelených a černých čajích a ve vzorcích instantních granulovaných čajů. Přítomnost těchto látek byla ověřena pomocí HPLC-MS. U všech vzorků byla změřena antioxidační kapacita metodou DPPH a ABTS a získané výsledky byly porovnány s množstvím antioxidantů ve vzorcích. |

Determination of derivates of flavone and methylxanthine in the beverage with tea extract was topic of this diploma thesis. The separation conditions of liquid chromatography method with UV/VIS detection as well as preparation of the sample were optimized. Using the optimized method, derivates of flavone and methylxanthine were determined in ice tea samples containing green or black tea extract, packet green and black tea samples and instant granulated tea samples. The content of the tested compounds in the samples were verified by HPLC-MS method. The antioxidant capacities of the tea samples were determined by method DPPH and ABTS and the obtained results were compared with the amount of antioxidants. |
