Výskyt arkobakterů a jejich přežívání v přítomnosti vybraných přírodních látek

dc.description.abstract	Bakterie rodu Arcobacter mohou způsobit nepříjemná onemocnění nejen lidí, ale i zvířat, a proto je třeba jejich výskyt v potravinách eliminovat. V této studii byly testovány vzorky potravin a vod na přítomnost arkobakterů a prováděna jejich identifikace metodou multiplex PCR. Dále bylo ověřeno přežívání těchto mikroorganismů v přítomnosti extraktů z rostlinných matric difúzní metodou v jamce a následně byla stanovena minimální inhibiční koncentrace pro jednotlivé extrakty. Pro experimenty byly zvoleny kmeny Arcobacter cryaerophilus a Arcobacter butzleri, a to jak sbírkové kmeny, tak izoláty získané z potravin. Všechny testované extrakty z přírodních látek působily na bakterie rodu Arcobacter inhibičně. Mezi jednotlivými testovanými kmeny byly nepatrné odlišnosti, ale jednoznačně nejúčinnější proti arkobakterům byly rakytníkové extrakty. Pro ověření inhibičního účinku tohoto extraktu na bakterie rodu Arcobacter byl proveden pokus v reálné potravině.	cze

dc.description.abstract-translated	Bacteria of the genus Arcobacter can cause unpleasant diseases in humans and animals, therefore their occurrence in foodstuffs must be eliminated. Samples of food and different types of water were tested for an occurrence of arcobacters in this study. Their identification was carried out by using multiplex PCR. Resistance of the genus Arcobacter against extracts from natural substances was evaluated by diffusion method. Finally, the minimum inhibitory concentration of extracts was determined for some arcobacters. Arcobacter butzleri and Arcobacter cryaerophilus have been chosen for the experiments (collection strains and isolates obtained from foodstuffs). All tested extracts from natural substances have caused inhibition of arcobacters. There were slight differences among the various tested strains. Clearly, the most effective were extracts from Sea buckthorn. To verify the inhibitory effect on bacteria of the genus Arcobacter the experiment in the real food was attempted.	eng
