dc.description.abstract |
Kofein je nejvíce užívanou psychoaktivní drogou na světě. Je to purinový alkaloid, řadící se do třídy derivátů methylxantinu. Nachází se v celé řadě rostlin, kde slouží pravděpodobně jako ochrana proti škůdcům. V lidském organismu je kofein metabolizován v játrech, kde je demethylován na paraxantin, theobromin a theofylin. Kofein je konzumován především pro svůj povzbuzující efekt, který působí napříč celým organismem. Jeho hlavním účinkem je antagonismus adenosinových receptorů. Dalšími účinky jsou inhibice fosfodiesterázy, mobilizace intracelulárního vápníku ze sarkoplazmatického retikula nebo interakce s GABAA receptory. Kofein má také neuroprotektivní účinky a může být využit při léčbě rakoviny. Sportovci využívají jeho prokazatelný ergogenní účinek. Toxicita kofeinu je nízká, smrtelná dávka se pohybuje okolo 150 mg na kilogram váhy. Kofein je tedy obecně považován za bezpečný. |

Caffeine is the most commonly used psychoactive drug in the world. It is a purine alkaloid, ranking class of methylxanthine derivatives. It is synthesized in a variety of plants, where probably serves as a protection against pests. In the human body, caffeine is metabolized in the liver through demethylation to paraxanthine, theobromine and theophylline. Caffeine is consumed mainly for its stimulant effect. The mechanism of the effect is the antagonism on adenosine receptors. Other effects are the inhibition of phosphodiesterase, mobilization of intracellular calcium from sarcoplasmic reticulum, or interaction with GABAA receptors. Caffeine has also a neuroprotective effect and can be used in cancer treatment. Athletes use its proven ergogenic effect. The toxicity of caffeine is low, the lethal dose is approximately 150 mg per kilogram of weight. Therefore, caffeine is generally regarded as a safe coumpound. |
