dc.description.abstract |
První část diplomové práce byla zaměřena na přípravu DEEMM derivátů
aminokyselin analyzovaných pomocí RP-HPLC se spektrofotometrickou detekcí. Dále byly
aminokyseliny derivatizované s využitím FMOC-Cl činidla. Výsledné deriváty byly poté
rovněž analyzovány prostřednictvím RP-HPLC, tentokrát však se spektrofotometrickou
i fluorimetrickou detekcí. Poslední část práce se zabývá nejen přípravou PITC derivátů
22 aminokyselin, ale také optimalizací jejich separace pomocí RP-HPLC systému
se spektrofotometrickou detekcí. Optimalizovaná separace byla využita pro analýzu derivátů
aminokyselin v reálných vzorcích medovin, předem extrahovaných na LC-SCX patronkách. |

The first part of this thesis was focused on the preparation of DEEMM
derivatives of amino acid analyzed by RP-HPLC with spectrophotometric detection. Further,
the amino acids were derivatized using FMOC-Cl reagent. The resulting derivatives were also
analyzed by RP-HPLC with spectrophotometric and fluorimetric detection. The last part
of the thesis deals with the preparation of PITC derivatives of 22 amino acids and with
optimizing of their separation by RP-HPLC system with spectrophotometric detection.
Optimised separation was used for analysis of amino acid derivatives in real samples
of meads, previously extracted by LC-SCX columns.
