dc.description.abstract |
Pro pochopení vlastností a funkcí reálných biologických membrán byly použity různé modely fosfolipidových membrán (PLMs). Mechanismy transportních procesů a vlastností membrán byly studovány za použití planárních fosfolipidových dvojvrstev. Elektrochemické chování PLMs bylo objasňováno především za pomoci elektrochemické impedanční spektroskopie (EIS). EIS byla použita ke sledování tvorby, stability, a také transportních procesů přes stabilizované planární PLMs (p-PLMs). Modelové syntetické membrány byly charakterizovány pomocí mikroskopie atomárních sil (AFM) a EIS. Technika pro izolaci protoplastů z buněk ječmene byla přizpůsobena a optimalizována pro získání cytoplazmatické membrány, která obsahuje specifické transportní systémy. Získané reálné části membrán byly smíchány s lecitinem za tvorby membrán a následně byly charakterizovány za pomoci EIS. Transportní procesy kadmia, jakožto environmentálně
důležitého prvku, a fytochelatinu PC2 přes modelové a modifikované PLMs (s reálnými částmi z protoplastů) byly studovány a charakterizovány. Pro umožnění přenosu fytochelatinu PC2 byly
nejprve použity buňkou pronikající peptidy (transportan 10 a mastoparan X). Pro stanovení přepravovaných látek byla využita voltametrie s visící rtuťovou kapkovou elektrodou (HMDE)
nebo s pevnými amalgamovými elektrodami (SAE). |

Different models of phospholipid membranes (PLMs) have been used for better understanding of properties and of functionality of real biological membranes. Mechanisms of transporting processes and properties of membranes were investigated on planar phospholipid bilayers. Electrochemical behavior of PLMs was studied by electrochemical impedance spectroscopy (EIS). EIS was utilized
to monitor formation, stability, and transporting process across the supported planar PLMs (p-PLMs). Model synthetic membranes were characterized by atomic force microscopy (AFM) and by EIS. The technique for isolation of protoplasts from barley cells was adopted and optimized to obtain the cytoplasmic membranes containing specific transporting systems. The gained real parts of membranes were mixed with lecithin and they were characterized by EIS. Transporting processes of the environmentally important element cadmium and of phytochelatin PC2 across model and modified PLMs (in mixture with real parts of protoplasts) ware studied. At first, the cell penetrating peptides (transportan 10 and mastoparan X) were used for to enable transfer of phytochelatin PC2. Voltammetry with hanging mercury drop electrode (HMDE) or with solid amalgam electrodes
(SAEs) was utilized for detection of transported species. |
