Tato diplomová práce se zabývá přípravou monolitických kapilárních kolon a následnou modifikací jejich povrchu N-isopropylakrylamidem jako teplotně citlivým monomerem. Na připravených kolonách jsme charakterizovali vliv složení mobilní fáze a pracovní teploty na retenci nízkomolekulárních látek s různou polaritou. Pomocí matematického modelu jsme zjistili, že zatímco vyšší koncentrace acetonitrilu v mobilní fázi vede vždy ke snížení retence, vyšší pracovní teplota zvyšuje na připravených kolonách retenci pouze polárních látek. Dále jsme pomocí tohoto modelu předpověděli eluci nízkomolekulárních látek a porovnali ji s experimentálními daty. V budoucnu by tento typ monolitických stacionárních fází mohl najít uplatnění ve vícerozměrné chromatografické analýze. |

The aim of this work is to prepare and characterize monolithic stationary phases modified with N-isopropylacrylamide as a thermally responsive monomer. We have used two-step surface grafting and prepared five monolithic capillary columns with different degree of surface modification. An effect of mobile phase composition as well as an effect of working temperature on the elution of small molecules has been studied. According to developed mathematical model, with higher concentration of acetonitrile in the mobile phase, the elution of small molecules decreases. On the other hand, elevated working temperature increases retention of small polar molecules on prepared columns. Developed model has been used to predict elution of small molecules on prepared columns. Finally, thermally responsive monolithic stationary phases might be used in two-dimensional liquid chromatography at elevated temperature. |
