Tato bakalářská práce shrnuje základní poznatky o
diabetes mellitus II. typu a vlivu s tearoyl-CoA desaturázy na diabetem poškozený metabolismus lipidů. Zabývá se vznikem diabetu II. typu, jeho
patogenezí, diagnostikou a v závěru i léčbou a prevencí. Součástí onemocnění diabete s mellitus II. typu jsou i přidružená onemocnění jako je
např. hypertenze a metabolické poruchy, které tvoří komplexní problém celého tohoto onemocnění.
V druhé části práce pojednává o stearoyl-CoA desaturáze. Stearoyl-CoA desaturáza je enzym endoplazmatického retikula, který způsobuje vznik dvojné vazby mezi uhlíky řetězce nasycených mastných kyselin, tím i jejich detoxikaci a syntézu mononenasycených mastných kyselin. Stále probíhající studie potvrzují, že deficit s
tearoyl-CoA desaturázy by mohl sloužit jako regulátor složitého mechanismu lipogeneze. Dále bylo prokázáno, že tento deficit brání ukládání tukové tkáně a zvyšuje citlivost tkání k inzulínu. Tyto poznatky mohou v
budoucnu vést ke zlepšení léčby diabetu II. typu. |

This bachelor diploma thesis summarizes basic knowledge about diabetes mellitus type 2 and the influence of stearoyl-CoA desaturase on lipid metabolism damaged by diabetes. This thesis deals with the emergence of diabetes type 2, its pathogenesis and diagnosis, and in the end, its treatment and prevention. Diabetes mellitus type 2 is associated with diseases such as hypertension and metabolic disorders that add to the complexity of this disease.
Second part of the thesis discusses stearoyl-CoA desaturase. Stearoyl-CoA desaturase is an enzyme of endoplasmic reticulum, which enables the formation of double bonds between carbons in the chains of saturated fatty acids, hence their detoxification and the synthesis of monounsaturated fatty acids. Ongoing studies confirm that the deficit of stearoyl-CoA desaturase could serve as a regulator of the complex mechanism of lipogenesis. It was further shown that this deficit precludes storage of adipose tissue and increases tissue sensitivity to insulin. These findings may lead to improvement in treatment of diabetes type 2 in the future. |
