Buněčné kultury izolovaných hepatocytů patří v dnešní době mezi nejpoužívanější biologické modely in vitro. Jsou používány při studiu buněčných mechanizmů specifických pro jaterní buňky, studiu jaterních onemocnění, hepatotoxicity a metabolizmu. Cílem práce bylo nejprve provést optimalizaci metod pro zjištění viability buněk kultivovaných na vícejamkových destičkách: stanovení aktivity laktátdehydrogenázy (LDH) a testu WST-1. Dalším cílem bylo pomocí těchto optimalizovaných metod popsat vliv změny kultivačních podmínek (denzita buněk, objem kultivačního média, doba seedingu) na viabilitu buněk, a to u intaktních buněk a u buněk inkubovaných s modelovou hepatotoxickou látkou - tBHP. Ze získaných dat byly určeny optimální podmínky stanovení WST-1 (37 °C; 1 hod.) a LDH (37 °C; 30 000 bb/j). Za těchto podmínek byl nejprve sledován vliv objemu kultivačního média na viabilitu buněk testem WST-1. Ze získaných hodnot bylo zjištěno, že objem média statisticky významně ovlivňuje viabilitu buněk - s rostoucím objemem se viabilita zvyšuje. Dále byl sledován vliv doby seadingu na viabilitu hepatocytů pomocí testu WST-1. Z výsledků vyplývá, že s rostoucí dobou seedingu se viabilita hepatocytů výrazně zvyšuje. U hepatocytů, které byly inkubovány s tBHP, byl vliv objemu média na viabilitu nejprve testován pomocí WST-1. Viabilita hepatocytů se snižovala s rostoucí koncentrací tBHP, rozdíl ve viabilitě mezi buňkami inkubovanými v 100 a 150 ?l (média a tBHP) ale nebyl statisticky významný. Působení tBHP bylo sledováno také pomocí stanovení intra- a extracelulární aktivity LDH, popř. LDHleakage. U extracelulární aktivity LDH a LDH-L byla prokázána toxicita tBHP vyšší u buněk inkubovaných ve 100 ?l v porovnání s buňkami inkubovanými ve 150 ?l. U intracelulární aktivity LDH nebyl ve viabilitě buněk nalezen žádný rozdíl. |

Hepatocyte cultures are the most used biological models in vitro currently. The cultures have been used to study of cell mechanisms which are specific for hepatocytes, as well as to study of liver diseases, hepatotoxicity and metabolism. The first aim of the thesis was to optimize the methods estimating the viability of cultured cells - lactate dehydrogenase assay (LDH) and WST-1 test. Further aim was to describe an influence of various cultivation conditions (cell density, cultivation medium volume, and seeding time) on the cell viability, both in intact cells and in tBHP treated cells. (tBHP was used as a hepatotoxin.) We determined the optimal conditions of WST-1 assay (37 °C; 1 h) and LDH assay (37 °C; 30 000 cells/well). These assays were used to the estimation of the influence of cultivation medium volume on the cell viability. We found that the volume of the medium influenced the viability significantly, i.e. the enhanced medium volume caused increase of the viability. As well, we tested the influence of the seeding time on the hepatocyte viability using WST-1 test. The results showed that the extended time of seeding caused greater cell viability. In tBHP treated rat hepatocytes, we assessed the influence of medium volume and time of seeding as well. We found that the viability was decreasing regarding the increasing tBHP concentration. However, the difference between the viability of the cells incubated in 100 and 150 ?l was insignificant. The tBHP treatment was characterized using intra- and extracellular LDH activity assay and LDH-leakage too. As results from extracellular LDH activity and LDH-L, greater toxicity was proved in cells treated in the volume of 100 ?l compared to the cells treated in 150 ?l. On the other hand, no difference was found in intracellular LDH activities. |
