Staphylococcus aureus je významným původcem alimentárních intoxikací. Tyto intoxikace jsou způsobeny enterotoxiny, které Staphylococcus aureus produkuje. V naší práci je zkoumán vliv kultivačního média, teploty a pH na produkci stafylokokových enterotoxinů. K experimentu byly použity produkční kmeny Staphylococcus aureus, u kterých bylo metodou PCR ověřeno, že obsahují gen pro produkci enterotoxinů B nebo H. Za optimálních podmínek byla provedena kultivace kmenů Staphylococcus aureus a vyprodukované SE byly přečištěny pomocí TRICIN-SDS-PAGEelektroforézy s následnou imunodetekcí - Western Blotting. Detekční limity pro tuto metodu byly stanoveny pro SEB na 0,1 ?g/ml vzorku, pro SEH na 5,6 ?g/ml. Byla také provedena metoda RPLA pro ověření produkce SEB a porovnána s výsledky Western blottingu. Vliv pH byl sledován s použitím kyseliny chlorovodíkové, mléčné a octové a hydroxidu sodného. Vliv teploty byl sledován v rozmezí 5 - 50 °C. Pro vliv kultivačního média byla použita kultivace v BHI bujónu a fyziologickém roztoku. Bylo zjištěno, že stafylokokový enterotoxin B byl produkován již při 5 °C, stafylokokový enterotoxin H až při teplotě pokojové (25 °C). Rozmezí pH pro produkci SEH i SEB bylo od 4.03 do 10.60. |

Staphylococcus aureus is a major etiological agens of alimentary intoxication. This intoxication is caused by staphylococcal enterotoxins. In our work the influence of culture medium, temperature and pH on the production of staphylococcal enterotoxins was studied. Production strains of Staphylococcus aureus that contained a gene for the production of enterotoxins B or H were used in our experiments. This was verified by PCR method. The cultivation of strains of Staphylococcus aureus was carried out under optimum conditions and produced SE were purified by TRICIN-SDS-PAGEelectrophoresis followed by imunodetection - Western Blotting. The detection limits established for this method were 0.1 ?g/ml for the SEB and 5.6 ?g/ml for the SEH. Also, RPLA method was performed to verify the SEB production and compare it with the results from Western blotting. The influence of pH was studied after the adjustment with hydrochloric acid, lactic acid, acetic acid and sodium hydroxide. Temperature effects were monitored in the range of 5 - 50 °C. The cultivations in BHI broth and solution saline were performed to study the influence of culture medium. It was found that the staphylococcal enterotoxins B were already produced at 5 °C and staphylococcal enterotoxins H at room temperature (25 °C). The range of pH for the production of SEH and SEB were from 4.03 to 10.60. |
