Diabetes mellitus 2. typu je závažné metabolické onemocnění, které má za následek řadu zdravotních komplikací. Jedna z nejzávažnějších je zhoršené hojení ran, které v mnoha případech může vést až k amputacím. Jako zvířecí model diabetu 2. typu byl použit potkan Zucker Diabetic Fatty (ZDF), který má mutaci leptinového receptoru. Tento potkan je novým modelem pro studium zhoršeného hojení ran vlivem diabetu. Naším cílem bylo zjistit metodou ELISA, jaké procesy se odehrávají v organismu potkanů během vývoje diabetu a stanovit, jak se v plazmě mění koncentrace bílkovin, které mají vliv na hojení. Dále je detekovat a lokalizovat v hojících se ranách pomocí imunohistochemie. Byly popsány měnící se hladiny glukózy, inzulínu, leptinu a inhibitoru plazminogenového aktivátoru (PAI-1) během vývoje diabetu. Tyto hladiny byly také zvýšeny u dospělých diabetických potkanů ve srovnání s kontrolami. Nebyly nalezeny významné rozdíly v koncentracích interleukinu-6 (IL-6) a fibrinogenu. Ve tkáni hojících se ran, odebrané v desátém dni, byl sledován IL-6, PAI-1 a fibrinogen. IL-6 byl ve velkém množství detekován převážně na povrchu diabetických ran a v okolí tukových buněk. Slabě pozitivní byly také fibroblasty. Nebyly nalezeny rozdíly v množství a lokalizaci u PAI-1 v kontrolních a diabetických ranách. Na PAI-1 byl pozitivní endotel kapilár a horní vrstva epitelu. Fibrinogen byl detekován ve formě fibrinové sítě na povrchu diabetických ran a v epitelu ran diabetických i kontrolních. Potkan ZDF je vhodný ke sledování mechanizmů zhoršeného hojení ran vlivem diabetu, ale není za daných podmínek vhodný ke sledování doprovodných změn diabetu, týkajících se plazmatických změn fibrinogenu a IL-6. |

Type II diabetes mellitus is a serious metabolic disease that is accompanied by a number of health complications. One of the most important is impaired wound healing, which in many cases may lead to limb amputation. Zucker Diabetic Fatty (ZDF) rat, which has a mutation in the leptin receptor, was used as an animal model for type II diabetes mellitus. It is a new rat model for studying the impaired wound repair. Our goal was to find out what processes take place in rats during the development of diabetes. ELISA method was used to determine plasma concentrations of selected proteins that play an important role in early wound healing. The second aim was to detect and locate the proteins in the experimental wounds using immunohistochemistry. Elevated levels of glucose, insulin, leptin and plasminogen activator inhibitor-1 (PAI-1) have been found in diabetic rats compared to the controls. No significant differences were found in the concentrations of interleukin-6 (IL-6) and fibrinogen. Time changes in glucose, insulin, leptin and PAI-1 levels during the development of diabetes have been reported. In the tissue, collected at day 10 after wounding, the presence of IL-6, PAI-1 and fibrinogen was monitored. IL-6 was detected in a large quantity mainly at the surface of diabetic wounds and around the adipose cells. The fibroblastic cells were weakly positive. No differences have been found in the quantity and localization of PAI-1 in control and diabetic wounds. The PAI-1 was positive in capillary endothelium and in the upper layer of the epithelium. Fibrinogen was detected in a form of the provisional matrix network in diabetic and control wounds. ZDF rat is suitable to monitor wound healing impaired due to presence of diabetes, but it is not suitable for monitoring the changes accompanying diabetes, that are related to plasma fibrinogen and IL-6 levels. |
