Fibrotizační pochody v játrech jsou důsledkem aktivace určitých typů buněk - jaterních hvězdicových buněk (HSC) a jaterních myofibroblastů (MFB). V této práci jsme se zabývali vlivem kultivačního prostředí na expresi genů v jaterních MFB potkanů. Srovnávali jsme běžný způsob kultivace buněk na polystyrenových Petriho miskách s kultivací v trojrozměrném (3D) prostředí a sledovali vliv dvou rozdílných látek - růstového faktoru FGF-1 a WY-14643, ligandu receptorů PPAR?. MFB byly izolovány opakovaným pasážováním z frakce neparenchymových jaterních buněk potkanů. Poté byly vysazeny na plast a do kolagenního, respektive fibrinového gelu. Do kultivačního média byly přidány sledované látky. Buňky byly sklízeny v různých časových intervalech. Poté z nich byla izolována celková RNA a syntetizována cDNA. Exprese genů byla stanovena pomocí rtRT-PCR. Data byla statisticky zpracována pomocí Studentova t-testu (p ? 0,05). Naše práce ukázala, že růstový faktor FGF-1 a ligand PPAR? WY-14643 ovlivňují expresi genů jaterních myofibroblastů. Účinek obou látek byl časově závislý. Mezi ovlivněnými geny byly proteiny extracelulární matrix, matrixové metaloproteinázy i některé cytokiny. Zjistili jsme, že je růstový faktor FGF-1 i syntetická látka WY-14643 rozdílně ovlivňují expresi mRNA v buňkách kultivovaných na plastové Petriho misce a v buňkách kultivovaných v 3D prostředí kolagenního gelu. |

Liver fibrogenesis is the result of activation of two cell types - hepatic stellate cells (HSC) and liver myofibroblasts (MFB). We have studied the influence of cellular environment on gene expression in rat liver MFB. We have compared the common way of cell cultivation on polystyrene Petri dishes with cell culture in threedimensional (3D) environment. The cells were treated with two different substances - fibroblast growth factor FGF-1 and the ligand of PPAR? receptors WY-14643. MFB were isolated from a fraction rat liver nonparenchymal cells by repeated passaging. They were plated on polystyrene culture dishes or in colagen or fibrin gels. The studied substances were added into the cultivation medium. The cells were harvested at different time intervals. Total RNA was isolated from the cells and cDNA was sythetised. Gene expression was determined by rtRT-PCR. Data were statistically analysed by Student t-test (p ? 0,05). Our study showed that growth factor FGF-1 and PPAR? ligand WY-14643 affect gene expression in liver MFB. Among the genes affected there were extracellular matrix proteins, matrix metaloproteinases and some cytokines. We have found that both the growth factor FGF-1 and the synthetic substance WY-14643 differently affect mRNA expression in cells cultured on plastic substrate and in the 3-D structure of collagen gel. |
