Diplomová práce se zabývá analýzou silic s využitím mikroextrakčních technik a destilace s vodní parou. Pro analýzu extraktů byla využita plynová chromatografie s plamenově – ionizačním detektorem. Mikroextrakce tuhou fází, mikroextrakce jednou kapkou, mikroextrakce tuhou fází ve spojení s hydrodestilací, dynamická mikroextrakce kapalnou fází a destilace s vodní parou byly aplikovány u vzorku levanduly lékařské (Lavandula augustifolia), šalvěje lékařské (Salvia officinalis), máty peprné (Mentha piperita) a meduňky lékařské (Melissa officinalis). V teoretické části jsou popsány klasické extrakční techniky. Jsou zde také vysvětleny výhody, nevýhody a využití mikroextrakce tuhou fází a mikroextrakce kapalnou fází. V této části jsou také uvedeny vlastnosti a chemické složení silic, jejich výskyt, využití a vliv na lidské zdraví. V experimentální části jsou uvedeny pracovní postupy úpravy, extrakce a analýzy vzorku, popis zařízení a seznam použitých chemikálií. Výsledky a diskuze obsahují optimalizaci dynamické mikroextrakce kapalnou fází a mikroextrakce tuhou fází ve spojení s hydrodestilací. Bylo provedeno kvalitativní a kvantitativní vyhodnocení zastoupení jednotlivých složek silic v bylinách a na základě dosažených výsledků bylo provedeno porovnání všech použitých extrakčních metod. |

This thesis deals with the essentials oils analysis in the herbs. These essential oils were isolated by microextraction techniques and steam distillation. The gas chromatography was used for analysis of extracts. The solid phase microextraction, single drop microextraction, solid phase microextraction combined with hydrodistillation, dynamic liquid phase microextraction and steam distillation were applied for analysis of Lavander (Lavandula augustifolia), Salvia (Salvia officinalis), Peppermint (Mentha piperita) and Balm (Melissa officinalis) samples. There are described classical extraction techniques in the theoretical part. There are explained of advantages, disadvantages and applications of the solid phase microextraction and the liquid phase microextraction techniques. The properties and chemical composition of essential oils, their presence, utilization and their influence on human’s health are described as well. There are introduced working processes, preparations, extractions and analysis of samples, descriptions of apparatuses and list of used chemicals in the experimental part. Results and discussion part involves optimization of dynamic liquid phase microextraction and solid phase microextraction combined with hydrodistillation. The qualitative and quantitative composition of essential oils in herbs and comparison of individual extraction methods were evaluated as well. |
