Oxokyseliny s rozvětveným řetězcem (ketoisovalerát, ketoisokaproát, ketomethylvalerát) jsou prvními produkty metabolismu při odbourávání aminokyselin s rozvětveným řetězcem. Vysoké hladiny oxokyselin s rozvětveným řetězcem v plazmě a krvi můžou způsobit nemoc javorového sirupu. Byla vyvinuta HPLC metoda s fluorescenční detekcí pro stanovení oxokyselin s rozvětveným řetězcem v plazmě a suchých krevních skvrnách. Oxokyseliny byly derivatizovány s o fenylendiaminem. Pro separaci derivátů byla použita chromatografická kolona LiChroCart 125-4 Purospher RP-18e, 5 ?m. Eluce byla provedena v gradientovém módu, kde jako mobilní fáze A byla použita směs methanolu s deionizovanou vodou (55:45, v/v pro plazmu a 50:50, v/v pro suché krevní skvrny) a mobilní fáze B obsahovala 100% methanol. Analytické parametry prezentované metody byly následující: přesnost v sérii, vyjádřená variačním koeficientem, byla v suchých krevních skvnách 3,67% pro KIV, 3,58% pro KIC a 3,18% pro KMV. Kalibrační křivka byla lineární v testovaném rozsahu koncentrací. Správnost vyjádřená výtěžností metody pro suché krevní skvrny KIV, KIC a KMV byla následující: 104,98% (CV 1,79%), 104,00% (CV 3,59%) a 101,99% (CV 4,85%). Vyvinuli jsme jednoduchou a selektivní HPLC metodu s fluorescenční detekcí pro stanovení oxokyselin v suchých krevních skvrnách a v plazmě. |

The branched-chain keto acids (ketoisovaleric acid, ketoisocaproic acid and ketomethylvaleric acid) are the first degradation products of these amino acids produced by enzymatic transamination with branched-chain amino acids transaminase. Highly increased concentrations of branched-chain keto acids in human plasma and blood lead to the clinical syndrome of maple syrup urine disease. A HPLC method with fluorescence detection for the measurement of branched-chain keto acids in plasma and dried blood spots has been developed and evaluated. The branched-chain keto acids were derivatized with o-phenylenediamine into 2 quinoxalinol derivatives. The quinoxalinol derivatives were separated by reversed-phase high-performance liquid chromatography using a column LiChroCart 125 4 Purospher RP-18e, 5 ?m with gradient elution. As mobile phase A was used the mixture of methanol and deionized water (55:45, v/v for plasma and 50:50, v/v for dried blood spots) and mobile phase B was used 100% methanol. The analytical performance of the method is satisfactory for clinical trials. The intra-assay coefficients of variation for KIV, KIC, KMV were 3.67%, 3.58% and 3.18% for dried blood spots. The calibration curve was linear with the tested range 0 100 ?mol/l for KIV, KMV, KIC. The recoveries were as follows: 104.98% (CV 1.79%), 104.00% (CV 3.59%) and 101.99% (CV 4.85%) for dried blood spots. We have developed a simple and a selective HPLC method with fluorescence detection for the determination branched-chain keto acids in dried blood spots and plasma. |
