| dc.description.abstract | Metabolic rewiring is often considered an adaptive pressure limiting metastasis formation; however, some nutrients available at distant organs may inherently promote metastatic growth. We find that the lung and liver are lipid-rich environments. Moreover, we observe that pre-metastatic niche formation increases palmitate availability only in the lung, whereas a high-fat diet increases it in both organs. In line with this, targeting palmitate processing inhibits breast cancer-derived lung metastasis formation. Mechanistically, breast cancer cells use palmitate to synthesize acetyl-CoA in a carnitine palmitoyltransferase 1a-dependent manner. Concomitantly, lysine acetyltransferase 2a expression is promoted by palmitate, linking the available acetyl-CoA to the acetylation of the nuclear factor-kappaB subunit p65. Deletion of lysine acetyltransferase 2a or carnitine palmitoyltransferase 1a reduces metastasis formation in lean and high-fat diet mice, and lung and liver metastases from patients with breast cancer show coexpression of both proteins. | dc.description.abstract-translated | Metabolické přepojení je často považováno za adaptivní tlak omezující tvorbu metastáz; Některé živiny dostupné ve vzdálených orgánech však mohou ze své podstaty podporovat metastatický růst. Zjistili jsme, že plíce a játra jsou prostředí bohatá na lipidy. Navíc pozorujeme, že premetastatická tvorba niky zvyšuje dostupnost palmitátu pouze v plicích, zatímco strava s vysokým obsahem tuku jej zvyšuje v obou orgánech. V souladu s tím cílené zpracování palmitátů inhibuje tvorbu plicních metastáz odvozených od rakoviny prsu. Mechanicky buňky rakoviny prsu používají palmitát k syntéze acetyl-CoA způsobem závislým na karnitin palmitoyltransferáze 1a. Současně je exprese lysin acetyltransferázy 2a podporována palmitátem, který spojuje dostupný acetyl-CoA s acetylací podjednotky jaderného faktoru-kappaB p65. Delece lysin acetyltransferázy 2a nebo karnitin palmitoyltransferázy 1a snižuje tvorbu metastáz u myší s chudou a vysokotučnou dietou a metastázy plic a jater u pacientek s karcinomem prsu vykazují koexpresi obou proteinů. Závěrem lze říci, že prostředí bohaté na palmitáty podporuje růst metastáz zvýšením acetylace p65, což vede k signalizaci prometastatického jaderného faktoru-kappaB. | cze |
