Tématem diplomové práce je stanovení vybraných aminokyselin v plazmě pacientů s diabetes mellitus. Byla popsána HPLC metoda s fluorescenční detekcí pro stanovení vybraných aminokyselin v lidské plazmě. Po odpovídající přípravě vzorku byly aminokyseliny derivatizovány s naftalen-2,3-dikarboxaldehydem za vzniku stabilního, vysoce fluoreskujícího derivátu. Pro separaci derivátů byla použita chromatografická kolona LiChroCart 250 x 4,0 mm Purospher Star RP-18e, 5 mikrom. Pro analýzu aminokyselin byla použita jako mobilní fáze A směs 25mmol/l NaH2PO4 a ethanolu (80:20, v/v), pH 7,2 ± 0,1, jako mobilní fáze B byl použit 100% ethanol. Analytické parametry metody u vybraných aminokyselin byly vyhovující. Linearita byla dosažena v celém rozsahu testovaných koncentrací. Přesnost v sérii vyjádřená variačním koeficientem byla pod 10 %. U pacientů s diabetes mellitus 2.typu byly v porovnání s kontrolní skupinou nalezeny statisticky významně vyšší hladiny aminokyselin s rozvětveným řetězcem. Vyvinuli jsme citlivou a dostatečně selektivní HPLC metodu s fluorescenční detekcí pro stanovení aminokyselin v lidské plazmě. |

The theme of the thesis is the determination of selected amino acids in plasma of patients with diabetes mellitus. A HPLC method with fluorescence detection for the measurement of selected amino acids in human plasma is described. After adequate sample preparation, the amino acids were derivatized with naphthalene-2,3-dicarboxaldehyde to form a stable, highly fluorescent derivative. For the separation, a reverse-phase column LiChroCart 250 x 4.0 mm, Purospher Star RP-18e, 5 mikrom, were used. For amino acids analysis, 25mmol/l sodium dihydrogenphosphate - ethanol (80:20, v/v), pH 7.2 ± 0.1 was used as a mobile phase A and 100% ethanol was used as a mobile phase B. The analytical parameters of this method were satisfactory for the determination of selected amino acids. The calibration curves were linear over the whole tested range. The intra-assay coefficients of variation were below 10%. In the patients with non-insulin dependent diabetes mellitus we found statistically significant higher levels of amino acids with branched chain compared to the control group. We developed a sensitive and selective high-performance liquid chromatography method with fluorescence detection for the determination selected amino acids in human plasma. |
