Rozvoj diabetu mellitu typu 2 (T2DM) je doprovázen dyslipidemickými procesy ovlivňujícími fyziologii různých tkání. Strukturální změny LDL částic jsou také známé pro svůj zvýšený aterogenní potenciál, proto se populace diabetiků druhého typu potýká se zvýšeným výskytem aterosklerózy a kardiovaskulárních chorob. Tato diplomová práce se zabývá složením mastných kyselin ve frakci LDL u pacientů s T2DM. Dále zkoumá závislost změny profilu mastných kyselin a aktivit desaturačních a elongačních enzymů na rozvoji tohoto onemocnění. Vzorky LDL frakcí od anonymních dárců byly nejprve analyzovány celkově na obsah mastných kyselin pomocí plynové chromatografie. Poté byl pro detailnější analýzu obsah mastných kyselin analyzován v jednotlivých lipidových třídách, na které byla frakce LDL separována pomocí tenkovrstevné chromatografie.
U pacientů s T2DM byly prokázány dyslipidemické změny, které se projevovaly nárůstem sérových TAG a poklesem HDL-C. Ačkoliv se koncentrace LDL-C od kontrolní skupiny nelišila, byl u pacientů prokázán pokles celkové koncentrace mastných kyselin a úbytek třídy esterů cholesterolu v LDL frakci. Dále byl u skupin pacientů zjištěn nárůst nasycených a pokles nenasycených a esenciálních mastných kyselin. Velmi výrazné rozdíly v porovnání kontrolní skupiny a pacientů vykazovala kyselina dokosahexaenová, která u pacientů klesala, a kyselina trans-vakcenová, jejíž koncentrace byla u pacientů s T2DM 100x vyšší. Byl také nalezen statisticky velmi významný vztah SCD-1 a glykovaného hemoglobinu. |

Development of diabetes mellitus type 2 (T2DM) is associated with dyslipidemia that affects physiological functions of different tissues. Structural changes of LDL particles are also known for their atherogenic potential, which is the reason for elevated incidence of atherosclerosis and cardiovascular diseases in type 2 diabetic population. This diploma thesis deals with composition of fatty acids in LDL fraction of T2DM patients. Furthermore, it studies a relationship of changes in profile of fatty acid composition in LDL and activity of desaturases and elongases on the development and onset of T2DM. Samples of LDL fractions from anonymous donors were firstly analyzed for overall composition of fatty acids by gas chromatography. Then the LDL fraction was separated into lipid classes by thin layer chromatography for more detailed analysis of fatty acids. We have demonstrated a dyslipidemia in patients with T2DM, that has manifested as elevated serum TAG and reduced levels of HDL cholesterol. Although, a cholesterol in LDL has not differed amongst groups of subjects, we have proved a reduced total concentration of fatty acids in LDL fraction and reduced levels of CE class. Furthermore, we have detected an increased levels of saturated fatty acids and reduced levels of unsaturated and essential fatty acids in LDL fraction. Both docosahexaenoic acid and trans-vaccenic acid have showed very significant differences in a comparison of patients to control group. Docosahexaenoic acid has had reduced levels in T2DM patients, meanwhile vaccenic acid has been 100 fold higher in these patients. Very strong relationship of SCD-1 and glycated hemoglobin has been also found. |
