dc.description.abstract |
Diplomová práce se zabývá detekcí nukleárních změn, které se objevují v průběhu apoptotického procesu. Teoretická část se věnuje popisu mechanismu apoptotické buněčné smrti s důrazem na jaderné změny, dále jsou zde popsány fluorescenční metody a fluorescenční sondy používané k detekci těchto jaderných změn. V závěru se teoretická část zabývá suspenzními buněčnými liniemi.
Experimentální část je zaměřena na detekci nukleární kondenzace a fragmentace pomocí kvantitativní spektrofluorimetrické metody Hoechst 33258 a především na její optimalizaci pro suspenzní buňky. Pro optimalizaci jsme zvolili buněčnou linii Jurkat, kterou jsme inkubovali s induktory apoptózy - cisplatinou, kamptotecinem a staurosporinem. Ke zhodnocení cytotoxicity vybraných toxinů jsme nejprve použili testy pro stanovení aktivity mitochondriálních dehydrogenáz a intracelulární hladiny glutathionu. Následně jsme detekovali nukleární kondenzaci a fragmentaci metodou Hoechst 33258 a provedli optimalizaci parametrů měření pro suspenzní buňky. Výsledky jsou shrnuty a diskutovány v závěru práce. |

The diploma thesis deals with the detection of nuclear changes that appear during the apoptotic process. The theoretical part is dedicated to description of the mechanism of apoptotic cell death with an emphasis on nuclear changes, and the fluorescence methods and fluorescent probes used to detect these nuclear changes are also described. Finally, the theoretical part deals with suspension cell lines.
The experimental part is focused on the detection of nuclear condensation and fragmentation using the quantitative spectrofluorimetric method Hoechst 33258 and especially on its optimization for suspension cells. For optimization, we chose the Jurkat cell line, which we incubated with apoptosis inducers - cisplatin, camptothecin and staurosporine. To evaluate the cytotoxicity of selected toxins, we first used tests to determine the activity of mitochondrial dehydrogenases and the intracellular level of glutathione. Subsequently, we detected nuclear condensation and fragmentation using the Hoechst 33258 method and optimized the measurement parameters for suspension cells. The results are summarized and discussed at the end of the thesis. |
