Tato práce přináší nové poznatky zdisoluční studie enterosolventně potahovaných tablet. Studie byla provedena na matricových tabletách připravených metodou přímého lisování s modelovou účinnou látkou. Tablety byly potahovány metodou 3D tisku. Testovány byly tablety hydrofilního charakteru (nosný polymer AffinisolTM) a lipofilního charakteru (na bázi Compritol? 888 ATO).
Jako potahový materiál byl použit AffinisolTM, AffinisolTM s přídavkem pektinu (5 hm. %) a PVA. Cílem studie bylo zjistit, zda potahované tablety odolají kyselému pH žaludku a jsou schopny zajistit transport léčiva do střeva. Z tohoto důvodu byla použita během disoluční studie média s pH 1,2 a 6,8, která odpovídají přirozenému prostředí v těle, a to žaludku a střevu. Disoluční testy probíhaly při teplotě 37 ? 0,5 °C a veškeré postupy byly v souladu s Českým lékopisem (Český lékopis 2017, Praha: Grada Publishing).
Získané disoluční profily, které popisují chování tablet při průchodu GIT, byly matematicky vyhodnoceny s použitím nelineární regresní analýzy. Výsledky jsou diskutovány zejména s ohledem na odolnost potahů v kyselém pH. |

The thesis presents new findings based on a dissolution study of enterosolvent- coated tablets. The study was conducted on matrix tablets prepared by direct compression with a model active ingredient. The tablets were coated by the 3D printing method. Tablets of hydrophilic nature (AffinisolTM carrier polymer) and lipophilic nature (based on Compritol? 888 ATO) were tested.
AffinisolTM, AffinisolTM with added pectin (5 hm. %) and PVA were used as coating material. The study aimed to determine whether the coated tablets can withstand the acidic pH of the stomach and are able to provide drug transport to the intestine. For this reason, media pH 1.2 and 6.8, which correspond to the natural environment in the body, namely the stomach and intestine, were used during the dissolution study. The dissolution tests were carried out at 37 ? 0,5 °C, and all procedures were in accordance with the Czech Pharmacopoeia.
The obtained dissolution profiles, which describe the behaviour of the tablets during passage through the GIT, were mathematically evaluated using non-linear regression analysis. The results are discussed especially with respect to the resistance of the coatings in acidic pH. |
