Moje diplomová práce se zabývala studiem enzymů cholinesteráz (acetylcholinesterázy a butyrylcholinesterázy) a jejich možných inhibitorů. Teoretická část této práce se zabývá popisem jejich struktury, funkce a významu v organismu. Dále se věnuje inhibici cholinesteráz, klasifikaci a vlastní struktuře různých inhibitorů. V teoretické části této práce jsou popsány metody stanovení anticholinergních aktivit inhibitorů a také je popis Schiffových bází, které budou v této práci experimentovány.
V experimentální části se tato práce zabývá stanovením aktivity enzymového preparátu (acetylcholinesterázy a butyrylcholinesterázy) a hodnot IC50 studovaných inhibitorů pomocí kolorimetrické Ellmanovy metody. Experimentální část je také věnována stanovení rozdělovacího koeficientu Kow studovaných inhibitorů v systému 1-oktanol:voda. |

My diploma thesis dealt with the study of cholinesterase enzymes (Acetylcholinesterase and Butyrylcholinesterase) and their possible inhibitors. The theoretical part of this work deals with the description of their structure, function and importance in the body. It also deals with cholinesterase inhibition, classification and structure of various inhibitors. In the theoretical part of this work are described methods of determination of anticholinergic activities of inhibitors and also the description of Schiff bases, which will be experimented in this work
In the experimental part, this work deals with the determination of the enzyme preparation activity (Acetylcholinesterase and Butyrylcholinesterase) and IC50 values of the studied inhibitors using the colorimetric Ellman´s method. The experimental part is also dedicated to the determination of the partition coefficient (P) of the studied inhibitors in the 1-octanol:water system. |
