Tato diplomová práce je součástí projektu zabývajícího se vývojem nového mikroprůtokového zařízení pro izolaci cirkulujících nádorových buněk z plné krve. Stanovení cirkulujících nádorových buněk v periferní krvi pacientů je důležité nejen pro včasné odhalení nádorového bujení, ale hlavně pro monitorování průběhu onemocnění, úspěšnosti protinádorové terapie a detekce návratu onemocnění. Předmětem naší práce bylo vyvinout bioaktivní nosič pro toto zařízení.
Cílem naší diplomové práce byl vývoj nového imunosorbentu s minimální nespecifickou sorpcí pro izolaci cirkulujících nádorových buněk a jeho porovnání s komerčně dodávaným nosičem. Ve výzkumné části jsme nejprve modifikovali povrch magnetických částic polyetylenglykolem, abychom dosáhli minimální nespecifické sorpce. Porovnávali jsme také účinnost vazby modelových IgG na magnetické částice PGMA povrchově modifikované polyetylenglykolem s funkčními skupinami karboxylovými, hydrazidovými nebo hydroxylovými. Dále jsme porovnávali nespecifickou sorpci a schopnost specifické vazby imobilizovaných protilátek povrchově modifikovaného imunosorbentu polyetylenglykolem s nosičem nemodifikovaným. Pro ověření účinnosti specifické vazby připraveného imunosorbentu jsme si zvolili jako modelový systém biospecifický pár protein p53 a protilátky anti-p53. Potom jsme porovnali specifickou vazbu buněk EpCAM+ a nespecifickou sorpci buněk EpCAM- při imunomagnetické separaci buněk námi připraveným imunosorbentem a komerčně dostupným imunosorbentem. Nakonec jsme porovnávali schopnost imunosorbentů izolovat buňky s různou mírou exprese molekuly EpCAM. Námi připravený imunosorbent se účinností specifické vazby buněk EpCAM+ vyrovnal komerčně dostupnému nosiči, ale nespecifická sorpce buněk EpCAM- byla vyšší. |

The graduation theses are part of a project dealing with development of a new micro-flow device for isolation of circulating tumour cells from full blood. The identification of circulating tumour cells in peripheral blood of patients is important not only for detection of tumour proliferation, but mainly for monitoring the disease process, the efficiency of an anti-tumour therapy and the detection of the disease recurrence. The subject of this work was to develop a bioactive carrier for the device.
The aim of the theses is to develop a new immunosorbent with minimal non-specific sorption for isolation of circulating tumour cells and its comparison with commercially supplied carriers.
At the beginning of the research stage, the surface of magnetic particles was modified using polyethyleneglycol in order to obtain the minimal non-specific sorption. Then we compared the efficiency of the model IgG bond to PGMA magnetic particles modified on the surface by polyethyleneglycol with functional carboxylic, hydrazidic or hydroxylic groups. Additionally, the non-specific sorption and the specific binding power of immobilized antibodies of the immunosorbent being modified on the surface by polyethyleneglycol were compared to a non-modified carrier. To validate the efficiency of the specific bond of the prepared immunosorbent, we chose the biospecific pair of p53 protein and anti-p53 antibody as a model system. Finally, we compared the specific bonds of cells and the non-specific sorption of EpCAM+cells using the immunosorbent which had been prepared by us and also the commercially available immunosorbent. In the end, the efficiency of the immunosorbents to isolate cells was compared to different rates of EpCAM molecule expression. The EpCAM specific binding efficiency of the immunosorbent which had been prepared by us was comparable with the commercially available carrier, but the non-specific sorption of the EpCAM- cells was higher. |
